11.2.2.2 Toggle SELEX

The aptamers selected by traditional SELEX have high binding affinities with typical dissociation constants (Kd) ranging from picomolar to nanomolar. Furthermore, the produced aptamers also show a high specificity for binding the target. Species cross-reactivity is often desired during the preclinical evaluation of a molecule in animal models, as being too specific may be a problem. There is also a Toggle SELEX method to generate species cross-reactive aptamers (Generation of Species Cross-reactive Aptamers Using “Toggle” SELEX). Rebekah White’s group described an in vitro selection strategy, called “toggling” SELEX, to generate aptamers that can bind both human and porcine thrombin. In their research, RNA aptamers that would be used both clinically in humans and testable in animal models were selected by “toggling” the protein target between human and porcine thrombin during alternating rounds of selection. After the “toggle” selection, a family of aptamers, which can bind human and porcine thrombin with high affinity simultaneously, can be yielded. If appropriate targets are available, the toggle strategy is a simple method to promote cross-reactivity. This strategy should facilitate the isolation of ligands with special cross-reactivity-requiring therapeutic and diagnostic applications (Fig. 11.3).
